Brunkow, Fred Ramsdell, and Shimon Sakaguchi have been fundamental to understanding the mechanisms of peripheral immune tolerance, transforming our understanding of the balance between immunity and self-tolerance. Their research identified the central role of the FOXP3 gene as a key regulator of regulatory T cells (Tregs) and demonstrated the essential role of this cell lineage in modulating immune responses.
